DAD CALCULATION WORKSPACE

Design the dose architecture.

Deterministic, traceable calculations for device-achievable doses, band-specific combination-product tolerances, capacity, and adjacent-dose overlap.

Create the Original Dose Design first. Dose overlap is calculated separately and does not influence initial dose generation.
Basic parameter levelChoose Fixed, Preferred (±45%), Flexible (±90%), or Free for every numeric parameter.

OPTIONAL SECOND STAGE

Dose-overlap optimization

▲

First review the Original Dose Design. Then optionally reduce and redistribute doses using one delivered-dose range.

Permitted minimum-dose endpoint change
%
Default 0%. Used only if moving the minimum retains a larger feasible dose count.
Permitted maximum-dose endpoint change
%
Default 0%. Used only if moving the maximum retains a larger feasible dose count.

Priority: maximize dose count; keep both endpoints unchanged; then keep minimum unchanged; then keep maximum unchanged; finally move both by the smallest permitted percentages. Multiple dose bands are optimized as one continuous system. Intermediate band maxima stay fixed as rule thresholds: an Actual Dose at or below a threshold uses the lower band, while a dose above it uses the higher band. Every dose and interface pair is therefore calculated with its applicable band's concentration, increment, device, drug, and ISO parameters. The minimum permitted optimized spacing is measured from the smallest adjacent Actual-Dose gap across the complete Original Dose Design, including band interfaces; a single percentage design uses its smallest original Actual-Dose percentage gap.

Band setupBand 1 is master; each later band starts as a copy and edits detach only the changed parameter.
Band target gridEach band generates its own Desired Doses from its minimum, maximum, count, and rule.
Actual DoseFeasible volume × that band’s drug concentration
Feasible volumeROUND((target dose ÷ concentration) ÷ increment) × increment
ISO transitionPT = α ÷ 0.05; variable-dose α is the band’s device increment
Total combination rangeLower/upper device volume × lower/upper drug concentration for Release or EoSL
Dose variation overlapMAX(0, MIN(upper A, upper B) − MAX(lower A, lower B))
Overlap percentageOverlap width ÷ (Actual Dose B − Actual Dose A) × 100; equal Actual doses report “-”.
Overlap optimizationCalculate searches feasible non-Fixed architecture candidates and minimizes the system maximum adjacent total-dose overlap across Release and EoSL; ties favor the closest requested design.
Interface ruleBoundary doses keep their own ranges; the higher dose band supplies the overlap acceptance limit.

ISO 11608-1 is treated as a minimum boundary. Better performance is allowed; worse performance is visibly flagged. This screening tool does not make a regulatory compliance determination.
